The gross conformation of protein-sodium dodecyl sulfate complexes.

چکیده
The interaction of sodium dodecyl sulfate with a wide variety of proteins is characterized by a high binding ratio when the monomer concentration of amphiphile exceeds 5 x 10W4 M. This binding ratio on a gram to gram basis is identical for all proteins investigated. The protein portion of the complex contains a high degree of order, and hydrodynamic studies suggest that the complex is a rodlike particle, the length of which varies uniquely with the molecular weight of the protein moiety. These results explain the empirical observation that proteins dissolved in aqueous solutions containing high concentrations of sodium dodecyl sulfate have electrophoretic mobilities on polyacrylamide gels which are a unique function of their molecular weights. In addition, the data suggest a possible model for the conformation of membrane proteins and their interactions with phospholipid.

Mobility of sodium dodecyl sulphate - protein complexes.
Reduced and unreduced lysozyme aggregates formed by formaldehyde cross-linking comprise a set of model compounds for studying the effects of protein conformation on the electrophoretic mobilities of sodium dodecyl sulphate-protein complexes. The reduced aggregates were indistinguisable from normal proteins, but the unreduced aggregates migrated anomalously fast by about 14%. Contrary to expecta...
متن کاملUnfolding and folding pathway of lysozyme induced by sodium dodecyl sulfate.
Proteins may exhibit an unfolding or folding state in the presence of a surfactant. In the present study, the unfolding and folding pathway of hen egg white lysozyme (HEWL) induced by sodium dodecyl sulfate (SDS) is studied. The stoichiometry obtained from isothermal titration calorimetry (ITC) provides guidelines for other techniques. The fluorescence spectra and circular dichroism show that t...
متن کاملPreparation of Sodium Dodecyl Sulfate Modified Pyrrolidine-1-dithiocarboxylic acid Ammonium Coated Magnetite Nanoparticles for Magnetic Solid Phase Extraction of Pb(II) from Water Samples
This paper describes the development of a procedure for Pb(II) ions removal from various water samples after magnetic solid phase extraction (MNPs) by magnetite nanoparticles (Fe3O4 NPs) modified with sodium dodecyl sulfate (SDS) and pyrrolidine-1-dithiocarboxylic acid ammonium (PDTCAA). The synthesis of Fe3O4 NPs was certified by characterization techniques including field emission scanning el...
متن کاملCrystallographic studies of protein denaturation and renaturation. 2. Sodium dodecyl sulfate induced structural changes in triclinic lysozyme.
Cross-linked triclinic lysozyme was denatured with sodium dodecyl sulfate. Removal of the denaturant resulted in a refolding of the protein to a conformation similar to but not identical with the native one. Three-dimensional x-ray diffraction data out to 3.2-A resolution were collected for two states in the refolding pathway, and appropriately weighted electron density difference maps were con...
